- Yes, definitely. I think that the regulation of mRNA stability (such as with the 5' UTR that Thomas mentioned today) is a really viable option for quick reporting. --dna1112 22:39, 6 Jun 2005 (EDT).
Also, you propose using temperature as the signal that will turn the switch's visual reporter off. If you take a look at the genetic toggle switch paper, they used temp controlled lambda repressor. The problem with this I think is that it is slow. Taking a look at Fig 4, it seems that it takes 3-5 hours to really turn off their GFP reporter. Since you say the UV input takes on the order of seconds, perhaps we can make or there exists a similar setup with light of different wavelength. Or perhaps there is a way to have 2 UV thresholds. I just mention this because the faster the system, the cooler it will be. Also it would be super-awesome if the cells could release a pigment although I haven't seen this done except with X-gal and I'm not sure how the Hello World group did there thing.
