Create Freezer Stock

This protocol specifies how to create a freezer stock from a BioBrick order. It assumes that the cells have been grown in culture as specified by the BioBrick to cell culture protocol.

  1. Pipet 1ml of cells into freezer tube (note: use the freezer tubes with the purple caps; the freezer tubes with the transparent caps will get stuck in the centrifuge).
  2. Centrifuge at 14k rpm for 1 min.
  3. Aspirate supernatant, retaining pellet.
  4. Add 0.4ml of LB + 20% glycerol solution. Resuspend cells.
  5. Freeze at -80C
