Cotransformations

Protocol for putting two plasmids into one set of cells. [We assume that we have two plasmids in AMP]

1. Find a BioBrick that is in Kan with an easy to resolve length (i.e. C0070).
2. Digest insert DNA with XbaI-PstI and vector DNA with XbaI-PstI. (See Digestion Protocol)
3. Cut out the part from the insert gel and the plasmid from the vector gel
4. Ligate.Transform.Culture.Miniprep.
5. Make plates that have both Kan and Amp.
