CaCl2 Solution

{The following protocol makes 1x CaCl2 solution for making cells competent.}

  1. Make 100mM PIPES Solution
  2. In a 500ml bottle, mix (w. stirrer):
    • 50ml 100mM PIPES soln
    • 375ml ddH2O
    • 4.41g CaCl2 (Sigma)
    • 75ml glycerol
      {Glycerol is highly viscous and best measured out using 25ml pipets}
  3. Stir well and vacuum sterilize.
  4. Keep on ice for immediate use; store @ 4
